Severely enlarged prostate?
check the stitching on the chamois and not just in the problem area - had a recent surprise sore spot and though all looked good in the problem area at the time a couple of weeks later found that the chamois was detached across the top rear - have you gained or lost weight? fit seems sensitive especially in already worn in shorts

Deterioration in the chamois in your shorts? Or not washing them everytime you use them? There's a lot of horrible germs down there - and your chammy could become 'caked', especially if you use creams. My bikes gets transported around a lot in the boot of my car and I have to lower the seat to get the frame in...therefore the seatpost is marked to make sure it's the correct height for me.
Funny thing is Ive been doing some specific streatches and foiund that my right side is alot less supple than the left!!
Would tilting my seat ever so slightly forward help?
Or even very slightly moving the seat forward so to have a more upright position?

Been using slipstream cream but going back to the tried and tested Assos or may even try veloskin as it gets great reports
Applying too much chamois cream can cause chaffing.
Same thing has happened to me on an 18month old bike. I changed the seat before christmas and have been fine until the last month or two - I've been blaming the newish seat + warmer weather but also sightly puzzled why it's suddenly such an issue.
Have a look at your hip mobility- they can stiffen with time if you are doing a lot of riding. If one side isn't moving as well as the other then that can cause chaffing on that side as your pelvis moves on the saddle to compensate for the lack of movement.
